• ELISA Kit Principle: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Folate Receptor 1, Adult (FOLR1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Folate Receptor 1, Adult (FOLR1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Folate Receptor 1, Adult (FOLR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Folate Receptor 1, Adult (FOLR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

• Description of Target: Folate receptor 1 adult is a protein that in humans is encoded by the FOLR1 gene. The protein encoded by this gene is a member of the folate receptor (FOLR) family. It is mapped to 11q13.4. FOLR1 expression in Jurkat cells facilitated MBG or EBO entry, and FR-blocking reagents inhibited infection of MBG or EBO. This gene product is a secreted protein that either anchors to membranes via a glycosyl-phosphatidylinositol linkage or exists in a soluble form. The FOLR1 gene encodes the adult folate receptor, or folate-binding protein (FBP), which has a high affinity for folic acid and for several reduced folic acid derivatives, and mediates delivery of 5-methyltetrahydrofolate to the interior of cells. FOLR1 is also an important regulator of milk protein synthesis.
