ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Sphingosine 1 Phosphate Receptor 5 (S1PR5). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Sphingosine 1 Phosphate Receptor 5 (S1PR5). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Sphingosine 1 Phosphate Receptor 5 (S1PR5), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Sphingosine 1 Phosphate Receptor 5 (S1PR5) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Description of TargetReceptor for the lysosphingolipid sphingosine 1-phosphate (S1P). S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues. Is coupled to both the G(i/0)alpha and G(12) subclass of heteromeric G-proteins. S1P activation on oligodendroglial cells modulates two distinct functional pathways mediating either process retraction or cell survival. S1P activation on O4-positive pre-oligodendrocytes induces process retraction via a Rho kinase/collapsin response-mediated protein signaling pathway. The S1P-induced survival of mature oligodendrocytes is mediated through a pertussis toxin-sensitive, Akt-dependent pathway. S1P activation on oligodendroglial cells modulates two distinct functional pathways mediating either process retraction or cell survival. These effects depend on the developmental stage of the cell.
