ELISA Kit Principle | The Aviva Systems Biology Tnf ELISA Kit (Mouse) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse Tumor necrosis factor in Plasma or Serum. This assay employs an antibody specific for Mouse Tnf coated on a 96-well plate. Standards and samples are pipetted into the wells and Tumor necrosis factor present in a sample is bound to the wells by the immobilized antibody. The wells are washed and anti-Tnf antibodies conjugated with horseradish peroxidase (HRP) are added. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Tnf bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm. The quantity of Tnf in the test sample can be interpolated from the standard curve constructed from the standards and corrected for sample dilution. | ||||||||||||||||||

Description of Target | Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation (By similarity). Induces insulin resistance in adipocytes via inhibition of insulin-induced IRS1 tyrosine phosphorylation and insulin-induced glucose uptake. Induces GKAP42 protein degradation in adipocytes which is partially responsible for TNF-induced insulin resistance (PubMed:25586176). Plays a role in angiogenesis by inducing VEGF production synergistically with IL1B and IL6 (By similarity) |
