ELISA Kit Principle | Aviva Systems Biology SSRP1 ELISA Kit (Human) (OKEI00246) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for SSRP1 has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and removed. A biotinylated detector antibody specific for SSRP1 is added, incubated and followed by washing. Avidin-Peroxidase Conjugate is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of a luminol substrate which is catalyzed by the HRP to produce light emission. The light emission is read by a luminometer (or photo-multiplier equipped instrument) and the intensity of the emitted light is proportional to the amount of sample SSRP1
ELISA Kit Range | 0.625 - 40 ng/mL
ELISA Kit Reproducibility | Intra-assay reproducibility was evaluated using 20 replicates of 3 samples representing low, middle and high level SSRP1. Inter-assay reproducibility was evaluated with 3 samples representing low, middle and high level SSRP1 using 8 replicates on each of 3 plates. Intra-Assay CV <10% Inter-Assay CV <12%

Protein Name | FACT complex subunit SSRP1 |
Description of Target | Component of the FACT complex, a general chromatin factor that acts to reorganize nucleosomes. The FACT complex is involved in multiple processes that require DNA as a template such as mRNA elongation, DNA replication and DNA repair. During transcription elongation the FACT complex acts as a histone chaperone that both destabilizes and restores nucleosomal structure. It facilitates the passage of RNA polymerase II and transcription by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, then subsequently promotes the reestablishment of the nucleosome following the passage of RNA polymerase II. The FACT complex is probably also involved in phosphorylation of 'Ser-392' of p53/TP53 via its association with CK2 (casein kinase II). Binds specifically to double-stranded DNA and at low levels to DNA modified by the antitumor agent cisplatin. May potentiate cisplatin-induced cell death by blocking replication and repair of modified DNA. Also acts as a transcriptional coactivator for p63/TP63. |
