:: | Background: SMAD7(Mother against decapentaplegic drosophila homolog of 7), also known as MADH7 or SMA- AND MAD-RELATED PROTEIN 7, is a protein that in humans is encoded by the SMAD7 gene. It belongs to the SMAD family of proteins, which belong to the TGFbeta superfamily of ligands. By somatic cell hybrid analysis, Topper et al.(1997) mapped the MADH7 gene to human chromosome 18. Topper et al.(1997) demonstrated that MADH7 and MADH6 could form complexes in endothelial cells. MADH7 was induced in cultured vascular endothelium by fluid mechanical forces and was capable of modulating endothelial gene expression in response to both humoral and biomechanical stimuli in vitro. By FISH, Roijer et al.(1998) refined the localization to 18q21.1. Lallemand et al.(2001) showed that cells stably expressing SMAD7 had increased susceptibility to apoptosis induced by TGFB, TNFA, serum withdrawal, or loss of cell adhesion. |
