ELISA Kit Principle | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to SET Domain Containing Protein 7 (SETD7). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to SET Domain Containing Protein 7 (SETD7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain SET Domain Containing Protein 7 (SETD7), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of SET Domain Containing Protein 7 (SETD7) in the samples is then determined by comparing the O.D. of the samples to the standard curve. | ||||||||||||||||||||||

Description of Target | SETD7 is a histone methyltransferase that specifically monomethylates 'Lys-4' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. It plays a central role in the transcriptional activation of genes such as collagenase or insulin. It is recruited by IPF1/PDX-1 to the insulin promoter, leading to activate transcription. It has also methyltransferase activity toward non-histone proteins such as p53/TP53, TAF10, and possibly TAF7 by recognizing and binding the.-.-K in substrate proteins. |
