Protocol InformationCitation: 1: Choi ES, Nichol JL, Hokom MM, Hornkohl AC, Hunt P. Platelets generated invitro from proplatelet-displaying human megakaryocytes are functional. Blood.1995 Jan 15;85(2):402-13. PubMed PMID: 7529062.
Experiment Name: Detection of Activation-Dependent Antigens on Platelets
Experiment Background: 1. P-selectin expression on thrombin-stimulated platelets.2. Fixed platelets were rinsed and incubated for I hour with MoAb against CD62 (Oxford, UK)
Experimental Steps: P-selectin. Culture- and plasma-derived platelets were washed by gel filtration." A set of platelets from each population was fixed immediately (resting platelets) with an equal volume of cold 1% paraformaldehyde, pH 7.4. for at least 2 hours. Another set of platelets was stimulated with 1 U/mL thrombin for 1 minute and then fixed as above (activated platelets). Fixed platelets were rinsed and incubated for I hour with MoAb against CD62 (Oxford, UK) or the isotype control mouse IgG, (Becton Dickinson) at final concentrations of 10 pglmL. Platelets were rinsed and incubated with goat anti-mouse-FITC (Cappel; 1:500) for 30 minutes, rinsed, and analyzed for fluorescence intensity by flow cytometry (FACScan Flow Cytometer, Becton Dickinson). The FACScan was calibrated using 2-um beads (Becton Dickinson).
Other Reagents Used: NaCI, KCI, MgCI2, glucose, HEPES, glutaraldehyde
