Description of Target The protein encoded by this gene is a member of a family of nuclear proteins related by the presence of a methyl-CpG binding domain (MBD). These proteins are capable of binding specifically to methylated DNA, and some members can also repress transcription from methylated gene promoters. This protein contains multiple domains: MBD at the N-terminus that functions both in binding to methylated DNA and in protein interactions; several CXXC-type zinc finger domains that mediate binding to non-methylated CpG dinucleotides; transcriptional repression domain (TRD) at the C-terminus that is involved in transcription repression and in protein interactions. Numerous alternatively spliced transcript variants encoding different isoforms have been noted for this gene.
Reconstitution and Storage Store at -20C. Avoid repeated freeze/thaw cycles.

Human U2OS cells
ChIP results obtained with the Aviva antibody directed against MBD1r. ChIP assays were performed using human osteosarcoma (U2OS) cells, the Aviva antibody against MBD1 and optimized PCR primer sets. Sheared chromatin from 1x10e6 cells and 1.5 ug of antibody were used per ChIP experiment. Beads only were used as a negative IP control. Quantitative PCR was performed with primers for the promoters of the MLH1 gene (used as a positive control) and CDC6 gene (used as a negative control). Figure shows the recovery, expressed as a % of input (the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).

Human MBD1
Determination of the antibody titer. To determine the titer of the antibody, an ELISA was performed using a serial dilution of the Aviva antibody directed against human MBD1, crude serum and Flow Through. The plates were coated with the peptide used for immunization of the rabbit. By plotting the absorbance against the antibody dilution, the titer of the purified antibody was estimated to be 1:20,000.

Human Hela cells
Western blot analysis using the Aviva antibody directed against MBD1r. Nuclear extracts of HeLa cells (40 ug) were analysed by Western blot using the Aviva antibody against MBD1 diluted 1:500 in TBS-Tween containing 5% skim milk. The position of the protein of interest is indicated on the right, the marker (in kDa) is shown on the left.
