Application InfoELISA: XMG1.2 monoclonal antibody can be used as a capture antibody in a sandwich ELISA for quantifying mouse IFN-Y protein levels. Biotinylated XMG1.2 antibody can be paired with a suitable purified capture antibody, with purified recombinant human IFN-Y as the standard. Western Blotting: The purified XMG1.2 antibody is also useful for Western blotting applications. For Western blotting, a concentration of 1-5ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig, OASB01897) has been found to enable detection of <= 100 ng/lane of recombinant mouse IFN-Y under reducing conditions. Immunofluorescence/Flow Cytometry: XMG1.2 antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IFN-Y -positive cells within mixed cell populations.4-6 Recombinant mouse IFN-Y or Rat IgG1 (OASB012111) should be used as a specificity control. FITC conjugated XMG1.2 antibodies are useful for intracellular staining in cells producing IFN-Y . Aviva intracellular staining buffers are recommended for this application. Neutralization: The LE/AF form of XMG1.2 can be used for neutralizing mouse IFN-Y bioactivity. A suitable LE/AF Rat IgG1 (OASB00095) should be used as an isotype control. Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.
Storage- The purified (UNLB) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of 100 mMborate buffered saline, pH 8.2. No preservatives or amine- containing buffer salts added. Store at 2- 8 C
- The fluorescein (FITC) conjugate is supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizingagent. Store at 2- 8 C
- The biotin (BIOT) conjugate is supplied as 0.5 mg labeled antibody in 1.0 mL of PBS/NaN3. Store at2- 8 C
- The low endotoxin, azide- free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mLof PBS. Aliquot and store at or below - 20 C
- Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide- free product at - 20 Cimmediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored asdirected.

Description of TargetThis gene encodes a soluble cytokine that is a member of the type II interferon class. The encoded protein is secreted by cells of both the innate and adaptive immune systems. The active protein is a homodimer that binds to the interferon gamma receptor which triggers a cellular response to viral and microbial infections. Mice deficient in this gene have increased susceptibility to viral, bacterial and parasitic infections and to several autoimmune diseases.
