ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha (PPARgC1a). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha (PPARgC1a). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha (PPARgC1a), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha (PPARgC1a) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Description of TargetTranscriptional coactivator for steroid receptors and nuclear receptors. Greatly increases the transcriptional activity of PPARG and thyroid hormone receptor on the uncoupling protein promoter. Can regulate key mitochondrial genes that contribute to the program of adaptive thermogenesis. Plays an essential role in metabolic reprogramming in response to dietary availability through coordination of the expression of a wide array of genes involved in glucose and fatty acid metabolism. Induces the expression of PERM1 in the skeletal muscle in an ESRRA-dependent manner. Also involved in the integration of the circadian rhythms and energy metabolism. Required for oscillatory expression of clock genes, such as ARNTL/BMAL1 and NR1D1, through the coactivation of RORA and RORC, and metabolic genes, such as PDK4 and PEPCK.
