ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Phospholipase A2, Group IIA (PLA2G2A). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Phospholipase A2, Group IIA (PLA2G2A). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Phospholipase A2, Group IIA (PLA2G2A), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Phospholipase A2, Group IIA (PLA2G2A) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ELISA Kit ReproducibilityIntra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Phospholipase A2, Group IIA (PLA2G2A) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Phospholipase A2, Group IIA (PLA2G2A) were tested on 3 different plates, 8 replicates in each plate. Gene Full Namephospholipase A2 group IIA
Alias Symbolseted enzyme type IIA phospholipase A2;GIIC sPLA2;group IIA phospholipase A2;phosphatidylcholine 2-acylhydrolase 2A;phospholipase A2, group IIA (platelets, synovial fluid);phospholipase A2, membrane associated;platelet phospholipase A2;sPLA2.
NCBI Gene Id29692
Protein NamePhospholipase A2, membrane associated
Description of TargetCatalyzes the calcium-dependent hydrolysis of the 2-acyl groups in 3-sn-phosphoglycerides (PubMed:3356705). Thought to participate in the regulation of phospholipid metabolism in biomembranes including eicosanoid biosynthesis. Acts as a ligand for integrins. Binds to and activates integrins ITGAV:ITGB3, ITGA4:ITGB1 and ITGA5:ITGB1. Independent of its catalytic activity, activates integrins by binding to a site (site 2) which is distinct from the classical ligand-binding site (site 1) and inducing integrin conformational changes and enhanced ligand binding to site 1. Induces cell proliferation in an integrin-dependent manner (By similarity).
