• ELISA Kit Principle: The microplate provided in this kit has been pre-coated with an antibody specific to Poly ADP Ribose Polymerase (PARP). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Poly ADP Ribose Polymerase (PARP). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Poly ADP Ribose Polymerase (PARP) level in the sample or standard.

• Description of Target: Involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks (PubMed:17177976, PubMed:18172500, PubMed:19344625, PubMed:19661379, PubMed:23230272).
