ELISA Kit Principle | Aviva's human OSM ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for OSM has been precoated onto 96-well plates. Standards(E.coli, A26R-220) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for OSM is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. Description of Target | OSM (Oncostatin M) is a protein that in humans is encoded by the OSM gene. OSM is a pleiotropic cytokine that belongs to the interleukin 6 group of cytokines. The OSM gene is mapped on 22q12.2. Of these cytokines it most closely resembles leukemia inhibitory factor (LIF) in both structure and function. Treatment with recombinant OSM leads to the inhibition of proliferation and changes in cellular morphology of a number of tumor cell lines derived from a wide variety of tissue types. OSM signals through cell surface receptors that contain the protein gp130. The type I receptor is composed of gp130 and LIFR, the type II receptor is composed of gp130 and OSMR. |
