20 ug of the indicated human whole cell extract was loaded onto a 4-20% SDS-PAGE gel. After electrophoresis, proteins were transferred to a PVDF membrane and probed with 3 ug/mL of antibody. Secondary detection was performed with 0.08 ug/mL of HRP-conjugated donkey anti-rabbit IgG antibody. Images were captured after development of membrane following exposure to AvivaBlot™ Femto ECL Reagent (OKCF00002).

Immunocytochemical analysis of ATM00002 was performed with 70% confluent PANC-1 cells. Cells were fixed and permeabilized with freeze-cold methanol for 20 min at -20C, blocked with 2.5% goat serum for 1hr, stained with ATM00002 at a 1:50 dilution in 2.5% goat serum, incubated overnight at 4C, and labeled with goat anti-Rabbit Alexa flour 488 (Jackson ImmunoResearch) at 1:1000 for 30 min at room temperature. Nuclei (DAPI) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938, Thermofisher Scientific). The images were capture at 40X magnification, Scale bars = 20 um.

Surface Plasmon Resonance Kinetic Characterization of ATM00002. Antibody was immobilized on a Protein A/G coated Carterra LSA sensor chip (PAGH200M) at concentrations of 5, and 50 ug/mL in duplicate. Antibodies on the surface were exposed to interaction with peptides sequentially via microfluidic controlled flow at 333nM peptide concentration for kinetic characterization of the binders for affinity and specificity, followed by curve fitting using the Kinetics software. Kd determinations for both concentrations were averaged and results and standard deviation are shown.

• Description of Target: This gene encodes a 105 kD protein which can undergo cotranslational processing by the 26S proteasome to produce a 50 kD protein. The 105 kD protein is a Rel protein-specific transcription inhibitor and the 50 kD protein is a DNA binding subunit of the NF-kappa-B (NFKB) protein complex. NFKB is a transcription regulator that is activated by various intra- and extra-cellular stimuli such as cytokines, oxidant-free radicals, ultraviolet irradiation, and bacterial or viral products. Activated NFKB translocates into the nucleus and stimulates the expression of genes involved in a wide variety of biological functions. Inappropriate activation of NFKB has been associated with a number of inflammatory diseases while persistent inhibition of NFKB leads to inappropriate immune cell development or delayed cell growth. NFKB is a critical regulator of the immediate-early response to viral infection.
