ELISA Kit PrincipleAviva Systems Biology Lipoteichoic acid ELISA Kit (OKEH02591) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Lipoteichoic acid antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Lipoteichoic acid and incubated. The Lipoteichoic acid found in the sample or standards competes with the biotinylated Lipoteichoic acid for limited binding sites on the immobilized anti-Lipoteichoic acid antibody. Excess unbound biotinylated Lipoteichoic acid and sample or standard Lipoteichoic acid is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Lipoteichoic acid captured in the well and inversely proportional to the amount of Lipoteichoic acid which was contained in the sample or standard.

Description of TargetLTA may bind to target cells non-specifically through membrane phospholipids, or specifically to CD14 and to Toll-like receptors. Binding to TLR-2 has shown to induce NF-kb expression (a central transcription factor), elevating expression of both pro- and anti-apoptotic genes. Its activation also induces mitogen-activated protein kinases (MAPK) activation along with Phosphoinositide 3-kinase activation. Lipoteichoic acid (LTA) is a major constituent of the cell wall of Gram-positive bacteria. These organisms have an inner (or cytoplasmic) membrane and, external to it, a thick (up to 80 nanometer) peptidoglycan layer. The structure of LTA varies between the different species of Gram positive bacteria and may contain long chains of ribitol or glycerol phosphate. LTA is anchored to the cell membrane via a glyceride. It acts as regulator of autolytic wall enzymes (muramidases). It has antigenic properties being able to stimulate specific immune response and is released from the bacterial cells mainly after bacteriolysis induced by lysozyme, cationic peptides from leucocytes, or beta-lactam antibiotics.
