ELISA Kit Principle | The Aviva Systems Biology LEP ELISA Kit (Human) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Leptin in Plasma or Serum. This assay employs an antibody specific for Human LEP coated on a 96-well plate. Standards and samples are pipetted into the wells and Leptin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and anti-LEP antibodies conjugated with horseradish peroxidase (HRP) are added. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of LEP bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm. The quantity of LEP in the test sample can be interpolated from the standard curve constructed from the standards and corrected for sample dilution. | ||||||||||||||||||

Description of Target | Key player in the regulation of energy balance and body weight control. Once released into the circulation, has central and peripheral effects by binding LEPR, found in many tissues, which results in the activation of several major signaling pathways (PubMed:17344214, PubMed:15899045, PubMed:19688109). In the hypothalamus, acts as an appetite-regulating factor that induces a decrease in food intake and an increase in energy consumption by inducing anorexinogenic factors and suppressing orexigenic neuropeptides, also regulates bone mass and secretion of hypothalamo-pituitary-adrenal hormones.
