ELISA Kit Principle | Aviva Systems Biology’s IL12B ELISA Kit (Mouse) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for IL12B has been pre-coated onto a 96-well plate. Standards or test samples are added to the wells, incubated and washed. A Biotin conjugated detector antibody specific for IL- Y is added, incubated and followed by washi. Next it is incubated with horseradish peroxidase (HRP)-Streptavidin, followed by wash. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample IL12B captured in well. | ||||||||||||||||||

Description of Target | IL-Y is a new member of the IL-12 heterodimeric cytokine family consists of IL-12p40 and IL-27p28 subunits, has important roles in regulating the immune response. IL-Y in particular is able to stimulate a specific subset of cytokines/chemokines, distinct from the IL-12 family members. It was reported that scIL-Y stimulated expression of MIP1α (CCL3), a chemokine involved in the recruitment of mononuclear cells, most notably neutrophils to the site of inflammation or injury and CCR5+ Treg cells. scIL-Y also specifically stimulated expression of the chemokine MIG (CXCL9), a T-cell chemoattractant, and the cytokines G-CSF, able to stimulate granulocytes and certain stem cell populations, IL-5 and IL-15. However, despite the ability of scIL-Y to stimulate expression of a specific subset of chemokines/cytokines, the secretion of endogenous IL-Y from stimulated mouse splenocytes is reported. This new ELISA Kits may be a useful tool to further investigate IL-Y. |
