ELISA Kit Principle | Aviva Systems Biology’s IL-10 ELISA Kit (Human) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for IL-10 has been pre-coated onto a 96-well plate. Standards or test samples are added to the wells, incubated and washed. A biotin conjugated detector antibody specific for IL-10 is added, incubated and washed. Next it is incubated with horseradish peroxidase (HRP)-Streptavidin, followed by wash. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample IL-10 captured in well. | ||||||||||||||||||

Description of Target | The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. |
