ELISA Kit Principle | Aviva Systems Biology Glutathione ELISA Kit (OKEH02622) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Glutathione antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Glutathione and incubated. The Glutathione found in the sample or standards competes with the biotinylated Glutathione for limited binding sites on the immobilized anti-Glutathione antibody. Excess unbound biotinylated Glutathione and sample or standard Glutathione is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Glutathione captured in the well and inversely proportional to the amount of Glutathione which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | Glutathione (GSH) is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. Thiol groups are reducing agents, existing at a concentration of approximately 5 mM in animal cells. Glutathione reduces disulfide bonds formed within cytoplasmic proteins to cysteines by serving as an electron donor. In the process, glutathione is converted to its oxidized form, glutathione disulfide (GSSG), also called L-(–)-glutathione. Once oxidized, glutathione can be reduced back by glutathione reductase, using NADPH as an electron donor. The ratio of reduced glutathione to oxidized glutathione within cells is often used as a measure of cellular toxicity. |
