Species: Rat Experiment Name: Immunostaining of tissues Experiment Background: HELEN et al. perfomed Immunostaining of tissues to analysis the Increased mononuclear cell migratory activity in tissues from normal rats treated with exogenous AGE-RSA. Experimental Steps: 1. After the i.v. injection of either unmodified RSA , AGE-RSA , or AGE-RSA plus aminoguanidine for 2 week, liver and kidney with 10% formaldehyde by standard procedures, paraffin-embedded tissue sections from all animal groups (n = 5 per group) were processed in triplicate for routine histological examination after staining with Harris' alum hematoxylin stain.2. The presence of monocytes / macrophages in tissues was determined by using the, macrophage-specific monoclonal antibody ED2 as primary antibody and biotinylated goat anti-mouse IgG [F(ab')2] (Cappel Research Products) as secondary antibody in 7 to 10-um sections of frozen tissues. Other Reagents Used: Acetone, chloroform Number Of Protocols: 1
