ELISA Kit Principle | Aviva Systems Biology CASR ELISA Kit (OKEH08149) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-CASR antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated CASR and incubated. The CASR found in the sample or standards competes with the biotinylated CASR for limited binding sites on the immobilized anti-CASR antibody. Excess unbound biotinylated CASR and sample or standard CASR is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated CASR captured in the well and inversely proportional to the amount of CASR which was contained in the sample or standard. | ||||||||||||||||||||||

Additional Information | Function: G-protein-coupled receptor that senses changes in the extracellular concentration of calcium ions and plays a key role in maintaining calcium homeostasis (PubMed:8255296). Senses fluctuations in the circulating calcium concentration and modulates the production of parathyroid hormone (PTH) in parathyroid glands (By similarity). The activity of this receptor is mediated by a G-protein that activates a phosphatidylinositol-calcium second messenger system (PubMed:8255296). The G-protein-coupled receptor activity is activated by a co-agonist mechanism: aromatic amino acids, such as Trp or Phe, act concertedly with divalent cations, such as calcium or magnesium, to achieve full receptor activation (By similarity). | ||||||||||||||||||||||
