ELISA Kit PrincipleAviva Systems Biology CASP3 ELISA Kit (Human) (OKCD06592) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for CASP3 has been pre-coated onto a 96-well plate (12 x 8 Well Strips) and blocked. Standards or test samples are added to the wells, incubated and removed. A biotinylated detector antibody specific for CASP3 is added, incubated and followed by washing. Avidin-Peroxidase Conjugate is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes to yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample CASP3 captured in the well.
ELISA Kit Range0.156-10ng/mL
ELISA Kit ReproducibilityIntra-assay reproducibility was evaluated using 20 replicates of 3 samples representing low, middle and high level CASP3. Inter-assay reproducibility was evaluated with 3 samples representing low, middle and high level CASP3 using 8 replicates on each of 3 plates.
Intra-Assay CV <10%
Inter-Assay CV <12%

Protein NameCaspase-3
Description of TargetCASP3 is a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 6, 7 and 9, and the protein itself is processed by caspases 8, 9 and 10. It is the predominant caspase involved in the cleavage of amyloid-beta 4A precursor protein, which is associated with neuronal death in Alzheimer's disease. Alternative splicing of this gene results in two transcript variants that encode the same protein.
