ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcium Channel, Voltage Dependent, Gamma Subunit 6 (CACNg6). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcium Channel, Voltage Dependent, Gamma Subunit 6 (CACNg6). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcium Channel, Voltage Dependent, Gamma Subunit 6 (CACNg6), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Calcium Channel, Voltage Dependent, Gamma Subunit 6 (CACNg6) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Description of TargetL-type calcium channels are composed of five subunits. The protein encoded by CACNG6 represents one of these subunits, gamma, and is one of several gamma subunit proteins. It is an integral membrane protein that is thought to stabilize the calcium channel in an inactive (closed) state. CACNG6 is a member of the neuronal calcium channel gamma subunit gene subfamily of the PMP-22/EMP/MP20 family and is located in a cluster with two similar gamma subunit-encoding genes.
