ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcium Channel, Voltage Dependent, L-Type, Alpha 1S Subunit (CACNa1S). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcium Channel, Voltage Dependent, L-Type, Alpha 1S Subunit (CACNa1S). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcium Channel, Voltage Dependent, L-Type, Alpha 1S Subunit (CACNa1S), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Calcium Channel, Voltage Dependent, L-Type, Alpha 1S Subunit (CACNa1S) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Description of TargetPore-forming, alpha-1S subunit of the voltage-gated calcium channel that gives rise to L-type calcium currents in skeletal muscle. Calcium channels containing the alpha-1S subunit play an important role in excitation-contraction coupling in skeletal muscle via their interaction with RYR1, which triggers Ca(2+) release from the sarcplasmic reticulum and ultimately results in muscle contraction. Long-lasting (L-type) calcium channels belong to the 'high-voltage activated' (HVA) group.
