20mM Tris (pH 7.4)
150 mM NaCl
2 mM EDTA
1% Triton X
10% glycerol
Primary antibody dilution and incubation time: 1:200, overnight at 4C and then 1 hour at room temp.
Secondary antibody used and dilution and incubation time: Donkey anti-rabbit HPRT, 1:2000, 1 hour room temp.
What controls were used in your experiment (positive/negative)? None
Please include your detailed WB Procedure/Protocol here: 1. Run 10% SDS-PAGE gel
2. Transfer to PVDF (1 hour )with the following buffer:
57.7g Glycine
12.1g Tris Base
800ml Methanol
bring volume up to 4L with dH2O
3. Block membrane 5% milk/ one hour room temp
4. Wash with 1X PBST
5. Incubate membrane overnight (4C) in antibody made in 2% BSA/PBST
6. Incubate membrane at room temp 1 hour in antibody
7. Wash 3X (15min each) with PBST at room temp
8. Incubate with rabbit secondary (1;2000)/ 2% milk/PBST, one hour room temp
9. Wash 3X (15min each) with PBST at room temp
10. Develop with ECL (Supersignal, Dura, Pierce)
