AvivaBlot™ Ultra Femto Competitor Analysis. Hela whole cell lysates were 2 fold serially diluted from 2.5 ug/lane to 0.039 ug/lane Total Protein. An Anti-HDAC1 antibody was used at 1:10,000, with a Goat anti-Rabbit IgG HRP used at 1:300,000. Exposure time of 120 seconds on an GE Imagequant LAS 4000 was used for image acquisition.

AvivaBlot™ Ultra Femto Signal to Noise Comparison. Triplicate blots containing 2 fold dilutions of HeLa cell lysate were incubated with Rabbit anti-Human HDAC1 1:15,000 and Goat anti-Rabbit IgG HRP 1:300,000 followed by imaging for 120 seconds with Imagequant LAS 4000 (GE Healthcare). Signal to noise measurements are shown for Utra Femto versus leading Competitor A.

Triplicate blots containing 2 fold dilutions of whole cell lysates were incubated with anti-HDAC1 (Human) at 1:15,000, and Goat anti-Rabbit-IgG HRP at 1:300,0000. Blots were imaged for 120 seconds with ImageQuant LAS 40000 (GE Healthcare). Integrated averaged signal intensity, showing linear response with amount of lysate over a wide range (DR = Dynamic Range, R2 = linearity).

  Mix 1:1 volume/volume prior to use

• Protocol Information:
  1. Perform electrophoresis, membrane transfer and antibody incubation and washes.
  2. Mix Ultra Femto Part A and Ultra Femto Part B 1:1 in a conical tube thoroughly. 7 mLs final is required for a standard mini blot (~64 cm2 ). Working solutions can be stored at 4° C for up to 3 days but should be warmed to room temperature before use.
  3. Apply Ultra Femto working solution at room temperature directly to semi-wet blot using 1 mL per 10 cm3 of membrane.
  4. Incubate 2 minutes at RT.
  5. Expose membrane using chemiluminescent imager or x-ray film. Use plastic wrap to avoid wetting film where appropriate.
  6. Exposure time is empirically determined, typical exposures require 60 to 180 seconds.

  AvivaBlot™ Ultra Femto ECL Reagent is a two component high performance enhanced chemiluminescent HRP substrate with an exceptional signal intensity and stable light output for low femtogram detection level. The formulation provides low background and a high signal to noise ratio. Available as sample size (2 X 5 mL) for a single blot application (100 cm2) or as 2 X 50 mL bottles or Part 1 and Part 2 reagent for 1000 cm2, sufficient for approximately 15 mini gel applications. The signal duration is 11 hours, and the reagent can be stored at room temperature for up to 1 year from date of receipt.
