AvivaBlot™ Pico One Competitor Analysis. Hela whole cell lysates were 2 fold serially diluted from from 5 ug/lane to 0.078 ug/lane Total Protein. An Anti-HDAC1 antibody was used at 1:2000, with a Goat anti-Rabbit IgG HRP used at 1:20,000. Exposure time of 120 seconds on an GE Imagequant LAS 4000 was used for image acquisition.

• Protocol Information:
  1. Perform electrophoresis, membrane transfer and antibody incubation and washes.
  2. Apply Pico One room temperature substrate directly to semi-wet blot using 1 mL per 10 cm3 of membrane. Standard sized mini blots are ~64 cm2 requiring ~7 mLs of solution.
  3. Incubate 2 minutes at RT.
  4. Expose membrane using chemiluminescent imager or x-ray film. Use plastic wrap to avoid wetting film where appropriate.
  5. Exposure time is empirically determined, typical exposures require 60 to 180 seconds.

  AvivaBlot™ Pico One ECL Reagent is an all in one enhanced chemiluminescent detection reagent, formulated pre-mixed and ready to use for immunoblot applications. Available as sample size (10 mL) for a single application (100 cm2) or as a 250 mL bottle for 2500 cm2, sufficient for approximately 35 mini gel applications. The signal duration is 3 hours, and the reagent can be stored at room temperature for up to 1 year from date of receipt. Primary antibodies are generally used at 1:100-1:5000 dilution range, and secondary antibodies from 1:1000 to 1:20,000 from 1 mg/mL stock solutions.
