ELISA Kit Principle | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT) in the samples is then determined by comparing the O.D. of the samples to the standard curve. | ||||||||||||||||||||||

ELISA Kit Reproducibility | Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT) were tested on 3 different plates, 8 replicates in each plate. Protein Name | Aryl hydrocarbon receptor nuclear translocator |
Description of Target | Required for activity of the Ah (dioxin) receptor. This protein is required for the ligand-binding subunit to translocate from the cytosol to the nucleus after ligand binding. The complex then initiates transcription of genes involved in the activation of PAH procarcinogens. The heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters and functions as a transcriptional regulator of the adaptive response to hypoxia (By similarity). The heterodimer ARNT:AHR binds to core DNA sequence 5'-TGCGTG-3' within the dioxin response element (DRE) of target gene promoters and activates their transcription (PubMed:28396409). |
