Description of TargetToll-like receptors (TLRs) play an essential role in the detection and elimination of invading pathogens. Recognition of microbial components by TLRs triggers activation of the innate immune system. Upon stimulation, these receptors interact with specific TIR domain-containing adaptor proteins. Five such adaptors have been discovered to date, MyD88, Trif (TIR-domain-containing adaptor inducing interferon-beta), TIRAP (TIR domain-containing adaptor protein)/Mal (MyD88 adaptor-like), TRAM (Trif-related adaptor molecule) and SARM (SAM and ARM-containing protein). MyD88 is common to all TLR-mediated pathways, which lead to the production of inflammatory cytokines, whereas TRIF mediates induction of IFN-beta in TLR3 and TLR4 signalling pathways. TIRAP/Mal is implicated in the TLR2- and TLR4-mediated MyD88-dependent signalling pathway. TRAM is specifically involved in the TLR4-mediated TRIF-dependent pathway. Thus, TIR domain-containing adaptors provide specificity of TLR signalling.
