ELISA Kit PrincipleThe test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Beta Adrenergic Receptor Kinase (BARK). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Beta Adrenergic Receptor Kinase (BARK). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Beta Adrenergic Receptor Kinase (BARK), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Beta Adrenergic Receptor Kinase (BARK) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ELISA Kit ReproducibilityIntra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Beta Adrenergic Receptor Kinase (BARK) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Beta Adrenergic Receptor Kinase (BARK) were tested on 3 different plates, 8 replicates in each plate. Gene Full NameG protein-coupled receptor kinase 2
Alias SymbolsADRBK1;adrenergic beta receptor kinase 1;BARK1;beta-adrenergic receptor kinase 1;beta-ARK-1;BETA-ARK1;G-protein coupled receptor kinase 2.
NCBI Gene Id156
Protein NameBeta-adrenergic receptor kinase 1
Description of TargetSpecifically phosphorylates the agonist-occupied form of the beta-adrenergic and closely related receptors, probably inducing a desensitization of them. Key regulator of LPAR1 signaling. Competes with RALA for binding to LPAR1 thus affecting the signaling properties of the receptor. Desensitizes LPAR1 and LPAR2 in a phosphorylation-independent manner (PubMed:19306925, PubMed:19715378). Positively regulates ciliary smoothened (SMO)-dependent Hedgehog (Hh) signaling pathway by faciltating the trafficking of SMO into the cilium and the stimulation of SMO activity (By similarity).
