ELISA Kit Principle | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to A Disintegrin And Metalloprotease 17 (ADAM17). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to A Disintegrin And Metalloprotease 17 (ADAM17). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain A Disintegrin And Metalloprotease 17 (ADAM17), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of A Disintegrin And Metalloprotease 17 (ADAM17) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
ELISA Kit Range | 0.156-10ng/mL
ELISA Kit Reproducibility | Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level A Disintegrin And Metalloprotease 17 (ADAM17) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level A Disintegrin And Metalloprotease 17 (ADAM17) were tested on 3 different plates, 8 replicates in each plate. Gene Full Name | a disintegrin and metallopeptidase domain 17 |
Alias Symbols | CD156b;disintegrin and metalloproteinase domain-containing protein 17;Tac;Tace;TNF-alpha convertase;TNF-alpha converting enzyme;TNF-alpha-converting enzyme. |
NCBI Gene Id | 11491 |
Protein Name | Disintegrin and metalloproteinase domain-containing protein 17 |
Description of Target | Cleaves the membrane-bound precursor of TNF-alpha to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Plays a role in the proteolytic processing of ACE2 (By similarity). Responsible for the proteolytic release of several other cell-surface proteins, including p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1 and the amyloid precursor protein. Acts as an activator of Notch pathway by mediating cleavage of Notch, generating the membrane-associated intermediate fragment called notch extracellular truncation (NEXT). |
