ELISA Kit Principle | Aviva Systems Biology AATF ELISA Kit (Human) (OKEI00135) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for AATF has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and removed. A biotinylated detector antibody specific for AATF is added, incubated and followed by washing. Avidin-Peroxidase Conjugate is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of a luminol substrate which is catalyzed by the HRP to produce light emission. The light emission is read by a luminometer (or photo-multiplier equipped instrument) and the intensity of the emitted light is proportional to the amount of sample AATF
ELISA Kit Range | 0.156 - 10 ng/mL
ELISA Kit Reproducibility | Intra-assay reproducibility was evaluated using 20 replicates of 3 samples representing low, middle and high level AATF. Inter-assay reproducibility was evaluated with 3 samples representing low, middle and high level AATF using 8 replicates on each of 3 plates. Intra-Assay CV <10% Inter-Assay CV <12%

Description of Target | May function as a general inhibitor of the histone deacetylase HDAC1. Binding to the pocket region of RB1 may displace HDAC1 from RB1/E2F complexes, leading to activation of E2F target genes and cell cycle progression. Conversely, displacement of HDAC1 from SP1 bound to the CDKN1A promoter leads to increased expression of this CDK inhibitor and blocks cell cycle progression. Also antagonizes PAWR mediated induction of aberrant amyloid peptide production in Alzheimer disease (presenile and senile dementia), although the molecular basis for this phenomenon has not been described to date. |
