- Description of Target:
- CCNB1 is a regulatory protein involved in mitosis. CCNB1 complexes with p34(cdc2) to form the maturation-promoting factor (MPF).The protein encoded by this gene is a regulatory protein involved in mitosis. The gene product complexes with p34(cdc2) to form the maturation-promoting factor (MPF). Two alternative transcripts have been found, a constitutively expressed transcript and a cell cycle-regulated transcript, that is expressed predominantly during G2/M phase. The different transcripts result from the use of alternate transcription initiation sites.

IP Protocol (Protein G coupled antibody):
Brain tissues were homogenized in a buffer containing 125 mM NaCl, 20mM Tris, pH7.4,
1mM MagCl2, 2% CHAP, 10% Glycerol, and protease inhibitor mirture.
Incubate on ice for 30 min.
Spin down 10 min at 14k rpm.
Add Protein G preclearance for 30 min on ice with rocking.
Spin down 5 min.
Add 6-12 of pre-coupled antibody-Protein G and rock 2 h on ice.
Spin down and wash 6 times.
Boil sample in sample buffer with beta-ME for 10 min.
Resolve on 4-20% Pierce Precise Protein gels.
Blot overnight with 2ug/ml antibodies.
Wash 2x7 min in TBS-tween.
Add secondary antibody (LiCor) for 1.5h.
Take images using LiCor.

|How was this sample prepared?||Cells were harvested from cell culture plates by scraping. Cell suspension was transferred into 1.5ml tubes and centrifuged for 2 minutes at 700g. After discarding the supernatant, cells were incubated with 150ul lysis buffer (containing 10% (v/v) Glycerol, 0.5% (v/v) NP-40, 0.1% (v/v) NaF, 1% (v/v) NaV and 0.2 % protease inhibitor cocktail) on ice. Susequently cells were sonified with 10% amplitude for 5 seconds. Samples were centrifuged for 10 minutes at 14,000 rpm in a common bench-top centrifuge and the supernatant was used for western blot analysis.|
|Primary antibody dilution and incubation time:||Primary antibody was diluted 1:5000 (in 5% dry milk in PBST) and incubated for 18 hours at 4 d C.|
|Secondary antibody used and dilution and incubation time:||Secondary antibody against rabbit-IgG (Pierce Biotechnology) was applied in a 1:5000 dilution and incubated for 1h.|
|What controls were used in your experiment (positive/negative)?||Untreated cells were used as a reference in this experiment.|
|Please include your detailed WB Procedure/Protocol here:||Proteins were blotted on a PVDF membrane. Thereafter the membrane was incubated in 5% milk and PBS-T (PBS+0.05% Tween20 (Roth)) at room temperature for 1h to block all unspecific protein binding sites. Specific primary and secondary antibodies were used for protein detection. Primary antibodies were diluted in 5% milk-PBS-T and incubated overnight at 4 d C on a roll mixer. After incubation the membrane was washed three times with 15 ml PBS-T for 10 minutes and specific secondary antibody against rabbit immunoglobulins was applied for 1h at room temperature.
The membrane was washed three times for 10 minutes in PBS-T to rinse off the extra secondary antibodies. The membrane was transferred onto a plastic bag and incubated with SuperSignal(R) West Pico Chemiluminescent Substrate, Thermo Scientific (1:1) for 1 minute before it was developed.
