RELA antibody - middle region (ARP30305_P050)

Description of Target:
p65 is a subunit of the nuclear factor kappa-B, a second messenger, which activates the transcription of a number of genes in multiple tissues. The inhibitory effect of I-kappa-B upon NF-kappa-B in the cytoplasm is exerted primarily through the interaction with p65. p65 shows a weak DNA-binding site which could contribute directly to DNA binding in the NF-kappa-B complex. NFKB1 (MIM 164011) or NFKB2 (MIM 164012) is bound to REL (MIM 164910), RELA, or RELB (MIM 604758) to form the NFKB complex. The p50 (NFKB1)/p65 (RELA) heterodimer is the most abundant form of NFKB. The NFKB complex is inhibited by I-kappa-B proteins (NFKBIA, MIM 164008 or NFKBIB, MIM 604495), which inactivate NFKB by trapping it in the cytoplasm. Phosphorylation of serine residues on the I-kappa-B proteins by kinases (IKBKA, MIM 600664, or IKBKB, MIM 603258) marks them for destruction via the ubiquitination pathway, thereby allowing activation of the NFKB complex. Activated NFKB complex translocates into the nucleus and binds DNA at kappa-B-binding motifs such as 5-prime GGGRNNYYCC 3-prime or 5-prime HGGARNYYCC 3-prime (where H is A, C, or T; R is an A or G purine; and Y is a C or T pyrimidine).[supplied by OMIM]. How do Aviva's reagents play a role in your experimental goals?

We are currently using some of the AVIVA antibodies for our projects on NFKappaB and Notch pathways.

What type of experimental sample are you using and how did you prepare it?

We have used colorectal cancer cell lines but soon will start with human tissue samples.

What controls were used in your experiment ? Please include your positive control.

To date we have performed Immuno fluorescence, Western blot and Chromatib IP, but we are planning to do IHC and endogenous IP in the near future. We have provided the conditions used in the reports that I already sent. I will also include them when I send new data.

How did you store the antibody after re-suspension?

Aliquoted at -20 degree C.

Have you used another antibody which has worked in your application?

In our testing, we compare your antibodies with the antibodies from Chemicon, Upstate and Santa Cruz that are the other companies we used to work with.

If the antibody works, do you plan to use it in future experiments or to publish your data? Why or why not?

We are planning to include your antibodies in our future publications.

Protocol: Western blot analysis of Ls174 colorectal cancer cells (parental cell line compared with cells transfected with an empty vector or a vector codifying for human hes1) using the indicated antibodies. Anti-hes1 was used at 1μg/ml in TBSt and incubated 0/N at 4 degrees. As secondary antibody we used a Goat anti-Rabbit HRP from DAKO.
