GRM6 is part of Group III which is linked to the inhibition of the cyclic AMP cascade.

How many ug’s of tissue/cell lysate run on the gel:
1: 10 ug WT mouse retina lysate
2: 10 ug GRM6 knockout mouse retina lysate
Primary antibody dilution: 1:1000
Secondary antibody: Anti-rabbit HRP
Secondary antibody dilution: 1:10000
Western Blot:
- Samples from retina lysates of wild type (wt) and nob3 (mGluR6 knock out) mice warmed 10’ at 37°C
- Loaded 10 ug/well
- Blocked 1 hour, room temperature, with 5% Milk-PBST (0.1% Tween)
- 1 hour incubation with primary antibodies diluted 1:1000 (1 ug/ml), room temperature, in 1% Milk-PBST
- 4 washes in PBST
- 1 hour incubation with secondary anti-rabbit-HRP conjugated antibody diluted 1:10000 (0.04 ug/ml), room temperature, in 1% Milk-PBST
- 4 washes in PBST
- Developed using SuperSignal West Femto Chemiluminescent Substrate diluted 1:5
- 30 seconds exposure
