• Description of Target: PTHLH is a member of the parathyroid hormone family. This hormone regulates endochondral bone development and epithelial-mesenchymal interactions during the formation of the mammary glands and teeth. This hormone is involved in lactation possibly by regulating the mobilization and transfer of calcium to the milk. The receptor of this hormone, PTHR1, is responsible for most cases of humoral hypercalcemia of malignancy. Four alternatively spliced transcript variants encoding two distinct isoforms have been observed.

How was this sample prepared?	Cell total protein was extracted with RIPA buffer. Samples were loaded with Laemli buffer + β-mercaptoethanol.
Primary antibody dilution and incubation time:	Anti-PTHrP at 1:250, 1:500, 1:1000, 1:2000 dilution; overnight incubation at 4 0C.
Secondary antibody used and dilution and incubation time:	Goat anti-rabbit, at 1:2000 dilution; 1h at room temperature.
What controls were used in your experiment (positive/negative)?	Positive: Rat Walker 256 tumor extract.
Please include your detailed WB Procedure/Protocol here:	1. Block the membrane for 1 hour at room temperature using 5% bovine serum albumin (BSA) in PBS/0.05% Tween. 2. Incubate membrane with 1:250-1:2000 dilutions of primary antibody ARP 33885 (initial concentration: 1µg/µl) in 5% BSA in PBS/0.05% Tween overnight at 4°C. 3. Wash the membrane (x3) with PBS/0.05% Tween, 5 minutes each. 4. Incubate the membrane with 1:2000 of peroxidase-conjugated anti-rabbit antibody in 5% BSA in PBS/0.05% Tween at room temperature for 1 hour. 5. Wash the membrane (x3) with PBS/0.05% Tween, 15 minutes each. 6. Develop membranes by ECL chemiluminescence following the manufacturer’s recommendations.
