  Description:
Cyclin H regulates CDK7, the catalytic subunit of the CDK- activating kinase (CAK) enzymatic complex. CAK activates the cyclin-associated kinases CDC2/CDK1, CDK2, CDK4 and CDK6 by threonine phosphorylation. CAK complexed to the core-TFIIH basal transcription factor activates RNA polymerase II by serine phosphorylation of the repetitive carboxyl-terminus domain (CTD) of its large subunit (POLR2A), allowing its escape from the promoter and elongation of the transcripts. It is involved in cell cycle control and in RNA transcription by RNA polymerase II. Its expression and activity are constant throughout the cell cycle.

Antigen Specificity:
Polyclonal antibody produced in rabbits immunized with a synthetic peptide corresponding to a region of human CCNH with an internal ID of P00328.
Reconstitution and Storage:
Antibody is lyophilized from PBS buffer with 2% sucrose. Add 100 µl of distilled water. Final antibody concentration is 1 mg/ml. For longer periods of storage, store at -20°C. Avoid repeat freeze-thaw cycles.
Key Reference:
Maekelae, T.P., et al., (1994) Nature 371:254-257.
Quality Control:
Rabbit Ig G is purified by Protein G/A affinity chromatography method.
Suggested starting concentrations are provided. Optimal dilutions should be determined by end-user. Differences in calculated versus apparent molecular weight may be due to post-translational modifications or protein hydrophobicity. For research use only not for diagnostic, human, or veterinary use.
Western Blot
Suggested dilution at 1µg/ml in 5% skim milk / PBS buffer, and HRP conjugated anti-Rabbit IgG should be diluted in 1: 50,000 - 100,000 as second antibody.
