  Description:
Cerebral deposition of amyloid beta peptide is an early and critical feature of Alzheimer's disease. Amyloid beta peptide is generated by proteolytic cleavage of amyloid precursor protein (APP) by two proteases, one of which is the protein encoded by BACE-1. The encoded protein, a member of the peptidase A1 protein family, is a type I integral membrane glycoprotein and aspartic protease that is found mainly in the Golgi. Four transcript variants encoding different isoforms of BACE-1 have been described. Human BACE-1 protein extra-cellular domain (from 28 AA to 393 AAB ) was expressed from E.coli and highly purified with its native enzyme activity. Purity: > 95% by SDS-PAGE. Protein concentration in package is 1mg/ml.
Species:
Human
Gene Symbol:
BACE-1
Gene Aliases:
ASP2, BACE, BACE1, FLJ90568, HSPC104, KIAA1149
Quality Control:
Substrate peptide based fluorescence assay. in buffer of 0.1 M sodium acetate, pH 4.5 at 37C with 5% DMSO. The increase of fluorescence (excitation at 350nm and emission at 490nm) was monitored during substrate hydrosis in a fluorescence spectrophotometer
Storage:
Liquid in 20 mM Tris, 20 mM KCl, 0.3 M NaCl, 10% glycerol, pH 7.8.. For long term storage aliquot and store at -4 °C.
Key Reference:
Beta-secretase cleavage of Alzheimer's amyloid precursor protein by the transmembrane aspartic protease BACE Science 286 (5440), 735-741 (1999)
Applications:
1:This recombinant proteinBACE-1 is an excellent protein for proteases functional assays; 2: highly active, so excellent for BACE-1 inhibitor screen assay.
Suggested starting concentrations are provided. Optimal dilutions should be determined by end-user. Differences in calculated versus apparent molecular weight may be due to post-translational modifications or protein hydrophobicity. For research use only not for diagnostic, human, or veterinary use.
