Product Description:Useful as a positive control for ChIP assays. Efficient fixation of a protein to chromatin in vivo is a crucial step for the Chromatin-immunoprecipitation (ChIP) technique. The extent of crosslinking is probably the most important parameter. Two major problems concerning the subsequent immunoprecipitation step should be taken into account: first, an excess of crosslinking can result in the loss of material or reduced antigen availability in chromatin, or both, and second, the relative sensitivity of the antigen epitopes to formaldehyde. For rapid checking chromatin IP efficiency. DNA from an aliquot of IP-chromatin solution can be first reverse crosslinked and purified by phenol extraction. Then, PCR reactions using specific primers to amplify certain known promoter regions can be performed to check whether the chromatin immunoprecipitation is successful:

The transcription factor NFkB is widely recognized as a critical mediator of immune and inflammatory responses. In most cell types, NFkB is found in the cytoplasm where it is associated with an inhibitory protein known as IkB. Upon a variety of stimuli (such as tumor necrosis factor treatment), NFkB will rapidly transfer to the nucleus by the induced phosphorylation and subsequent degradation of IkB. In the nucleus, NFkB regulates genes encoding cytokines, cytokine receptors, cell adhesion molecules, proteins involved in coagulation and genes involved in cell growth control. In this NFkB ChIP-PCR assay kit, three known NFkB binding promoters and three negative genomic targets were selected and validated for use as control system for rapidly mapping a fixation condition of a interested protein to chromatin in vivo.
