AviLight ® Chemiluminescent Substrate is a high-sensitivity enhanced substrate for detecting horseradish peroxidase (HRP) on immunoblots. AviLight's extremely intense signal output enables detection of antigen at the picogram level. The sensitivity, intensity and duration of the signal allow for easy detection of HRP using photographic or other CCD camera based imaging methods. Blots can also be repeatedly exposed to film to obtain optimal results or stripped for immunoblotting with other antibodies.

  Storage Conditions: Store at 4°C for up to 12 months.  
  Assay Procedure: Sample and antibody use may require optimization.
Recommended antibody dilutions must be used to obtain best results.
Dilute primary antibody to 0.2-1.0 µg/ml from Aviva Antibody stock.
Dilute secondary antibody to 1:50,000-1:100,000 dilution from a 1 mg/ml stock.
Mix the two substrate components A & B at a 1:1 ratio to prepare a fresh substrate Working Solution.
Incubate blot for 5 minutes in AviLight substrate working solution.
Drain excess reagent. Cover blot with clear plastic wrap.
Expose blot to X-ray film.

Product Description:Useful as a positive control for ChIP assays. Efficient fixation of a protein to chromatin in vivo is a crucial step for the Chromatin-immunoprecipitation (ChIP) technique. The extent of crosslinking is probably the most important parameter. Two major problems concerning the subsequent immunoprecipitation step should be taken into account: first, an excess of crosslinking can result in the loss of material or reduced antigen availability in chromatin, or both, and second, the relative sensitivity of the antigen epitopes to formaldehyde. For rapid checking chromatin IP efficiency. DNA from an aliquot of IP-chromatin solution can be first reverse crosslinked and purified by phenol extraction. Then, PCR reactions using specific primers to amplify certain known promoter regions can be performed to check whether the chromatin immunoprecipitation is successful:

In most cell types, NFkB is found in the cytoplasm where it is associated with an inhibitory protein known as IkB.
