Description of Target:
COPS5 is one of the eight subunits of COP9 signalosome, a highly conserved protein complex that functions as an important regulator in multiple signaling pathways. The structure and function of COP9 signalosome is similar to that of the 19S regulatory particle of 26S proteasome. COP9 signalosome has been shown to interact with SCF-type E3 ubiquitin ligases and act as a positive regulator of E3 ubiquitin ligases. This protein is reported to be involved in the degradation of cyclin-dependent kinase inhibitor CDKN1B/p27Kip1. It is also known to be an coactivator that increases the specificity of JUN/AP1 transcription factors. Reconstitution and Storage:
Add 50 μl of distilled water. Final anti-COPS5 antibody concentration is 1 mg/ml in PBS buffer. For longer periods of storage, store at -20°C. Avoid repeat freeze-thaw cycles.

Cell types are:
1) B8 mouse fibroblast stably cells expressing Flag Csn2 (“mB8”).
2) HEK293 stably cells expressing Flag Csn2.
a. “Crude” - extraction only.
b. “Pure” - Partially purification by anti Flag column.
3) CSN complex purified from Human erythrocytes blood cells (“hBlood”):
a. Partially purification (DEAE column follow by Ammonium Sulfate, Glycerol gradient then MonoQ).
b. Homogeneous Purification (as “2a” and final by ResourceQ column).
4) CSN complex purified from S-Pombe cells expressing TAP-Tag peptide:
a. Partially purification (DEAE column follow by Ammonium Sulfate then anti IgG column ).
b. Purified CSN complex (DEAE column follow by Ammonium Sulfate, anti IgG column, Calmodulin column then Sucrose gradient).

Samples preparation:

Cells cultures:
All cell cultures (B8 mouse cells and HEK293 human cells) were extracted in RIPA buffer (extraction conditions: 10E7 cells per 120ul of buffer). HEK 293 cells expressing Flag Csn2, have been used either as a crude extract or as an one step Flag-purified CSN comlpex, according to Pick E. et al. 2008 MCB.

CSN preparation from Human Red Blood Cells:
CSN complex have been purified from human blood cells according to Hetfeld BK et al. 2005 Methods Enzymol. Red Cells had precipitated from 0.5 liter blood. Partially purified complex samples had taken from 1.5 ml Mono Q (AEX) peak fractions, the purified samples had taken from 1ml Resource Q (AEX) peak fractions.

Purificatrion of CSN from S-Pombe:
CSN complex have been purified from S-Pombe cells as describe above (The complex purified by unpublished protocol) from 159 gr dry cells. “Crude” samples had taken from 1 ml of IgG column peak fractions and the “Pure” samples had taken from 1ml of Sucrose Gradient column peak fraction. None of the antibody happened to recognize S. Pombe ortholog CSN subunits.

SDS – Page and immune bloting:
Running: 40mA for each gel; Transfer: 200 mA (2 gels) for 1h 30’. Blocking: Long life milk 3% fat 1:10. Antibodies in TBST with 0.02% Sodium Azid.
