ELISA Kit Principle | Aviva's human Adiponectin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Adiponectin has been precoated onto 96-well plates. Standards(NS0, E19-N244) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Adiponectin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Adiponectin amount of sample captured in plate. | |||||||||||||||||||||||||||||||||||

The TMB Color Developing agent is colorless and transparent before using, contact us freely if it is not the case. 3. In order to avoid marginal effect of plate incubation due to temperature difference ( reaction may be stronger in the marginal wells), it is suggested that the diluted ABC and TMB solution will be pre-warmed in 37C for 30 min before using. Background: Adiponectin (ADPN) is a hormone secreted by adipocytes that regulates energy homeostasis and glucose and lipid metabolism.1 Adiponectin is a new member of the family of soluble defense collagens, in hematopoiesis and immune responses. It is an important negative regulator in hematopoiesis and immune systems and raise the possibility that it may be involved in ending inflammatory responses through its inhibitory functions.2 Adiponectin is mapped to 3q27 and can protect the organism from systemic inflammation by promoting the clearance of early apoptotic cells by macrophages through a receptor-dependent pathway involving calreticulin.3 The standard product used in this kit is the product of gene recombination, consisting of 226 (19-244) amino acids with the molecular mass of 36KDa after glycosylation. | |||||||||||||||||||||||||||||||||||
